Biological Background: FXR2 Function and Localization
- FXR2 (Fragile X mental retardation syndrome-related protein 2) is an RNA-binding protein belonging to the FXR family, which includes FMR1, FXR1, and FXR2. It is also known as FMR1 autosomal homolog 2.
- It functions as a regulator of mRNA translation and stability by specifically binding to AU-rich elements (AREs) in the 3'-UTR of target mRNAs (By similarity).
- FXR2 promotes the formation of phase-separated membraneless compartments through liquid-liquid phase separation upon binding ARE-containing mRNAs, thereby modulating mRNA storage and translation (By similarity).
- The protein is required for adult hippocampal neurogenesis, where it regulates the translation and/or stability of NOG (noggin) mRNA to prevent NOG protein expression in the dentate gyrus (By similarity).
- Subcellularly, FXR2 localizes to the cytoplasm, cytoplasmic ribonucleoprotein granules, and the postsynaptic region.
- It is widely expressed in tissues including heart, brain, kidney, and testis. In the brain, FXR2 is present at high levels in neurons, particularly in Purkinje cells at the interface between the granular and molecular layers. Related references: PMID:9259278
