Biological Background: FXR2 Function and Localization
- RNA-binding protein that acts as a regulator of mRNA translation and/or stability, and which is required for adult hippocampal neurogenesis.
- Specifically binds to AU-rich elements (AREs) in the 3'-UTR of target mRNAs.
- Promotes formation of phase-separated membraneless compartments by undergoing liquid-liquid phase separation upon binding to AREs-containing mRNAs, thereby regulating mRNA storage, translation and/or stability.
- Acts as a key regulator of adult hippocampal neurogenesis by controlling NOG mRNA translation/stability, preventing NOG protein expression in the dentate gyrus.
- Localizes to the cytoplasm, cytoplasmic ribonucleoprotein granules, and the postsynapse.
- Expressed in all tissues examined including heart, brain, kidney and testis. In brain, present at high level in neurons and especially in the Purkinje cells at the interface between the granular layer and the molecular layer. Related references: PMID:9259278
- Belongs to the FMR1 family and is also known as FMR1 autosomal homolog 2.
- Post-translational modifications include phosphorylation, as indicated by the Phosphoprotein keyword.
Experimental Guidance and Technical Tips
- For Western blot, expect a specific band around 74 kDa based on the predicted molecular weight. Validate the antibody in relevant cell or tissue lysates, such as neuronal cells or brain tissue.
- For immunofluorescence/ICC, use appropriate fixation and permeabilization protocols to detect cytoplasmic and granular localizations. Include proper negative controls to confirm specificity.
