Biological Background: FXR1 Function and Localization
- FXR1, an autosomal homolog of the fragile X mental retardation protein FMR1, is an mRNA-binding protein that regulates translation and stability of target transcripts, playing essential roles in neurogenesis, muscle development, and spermatogenesis. Related references: PMID:17382880, PMID:20417602, PMID:30067974
- It specifically recognizes AU-rich elements (AREs) in 3'-UTRs of mRNAs, such as MYC, and can recruit the eIF4F complex to enhance translation initiation. Related references: PMID:17382880, PMID:34731628
- Through liquid-liquid phase separation, FXR1 assembles mRNA into cytoplasmic ribonucleoprotein granules, a process critical for activating translation of stored mRNAs in late spermatids and for muscle tissue development.
- FXR1 localizes to the cytoplasm, stress granules, dendrites, dendritic spines, and axons, and exhibits high protein levels in neurons, especially in cerebellar Purkinje cells. Related references: PMID:7781595, PMID:9259278
- It acts as a negative regulator of inflammation by binding and destabilizing pro-inflammatory transcripts, including TNF mRNA, thereby limiting excessive immune responses. Related references: PMID:30067974
- FXR1 prevents ectopic phase separation and cytoplasmic aggregation of nucleoporins, ensuring proper nuclear pore complex localization, and it also scaffolds a cytoplasmic mRNP network to facilitate actomyosin-related signaling. Related references: PMID:32706158, PMID:39106863
Experimental Guidance and Technical Tips
- This rabbit monoclonal antibody was raised against the N-terminal region (aa 1-100) of human FXR1; consider verifying epitope conservation if working with species or isoforms beyond the validated human, mouse, rat.
- In western blot, the predominant band is expected near 70 kDa, but alternative splicing may produce additional isoforms; validate with appropriate positive and negative controls in your sample system.
- For immunoprecipitation, FXR1 forms ribonucleoprotein complexes; include RNase-free conditions and appropriate controls to distinguish direct from indirect interactions.
- In immunofluorescence/ICC, FXR1 can accumulate in cytoplasmic granules and neuronal projections; optimize fixation/permeabilization and consider co-staining with granule markers (e.g., G3BP1 for stress granules).
- The antibody’s cross-reactivity with mouse and rat enables translational studies across species; however, confirm reactivity in your specific tissue or cell type of interest.
- Given its RNA-binding function, avoid RNase contamination during sample preparation and staining to preserve FXR1-mRNA complexes and epitope accessibility.
