Biological Background: FUT8 Function and Localization
- FUT8, encoded by the FUT8 gene, is an alpha-(1,6)-fucosyltransferase also known as CDGF or CDGF1.
- It catalyzes the addition of fucose in alpha 1-6 linkage to the first GlcNAc residue of N-glycans, a critical modification of glycoproteins including the Fc region of immunoglobulins. Related references: PMID:17172260, PMID:29304374, PMID:36280670
- FUT8 is a Golgi-resident type II membrane protein, localizing specifically to the Golgi stack membrane.
- It belongs to the glycosyltransferase family and features a signal-anchor, a transmembrane helix, and an SH3-binding domain.
- Mutations in FUT8 are associated with congenital disorder of glycosylation (CDG), highlighting its clinical significance.
- The enzyme undergoes phosphorylation and alternative splicing, events that may modulate its activity or isoform expression.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 376-575 of human FUT8, targeting the C-terminal region; consider whether this region may be present in alternative splicing isoforms and validate detection in your sample system.
- For Western blot, a band near 67 kDa is expected; use appropriate positive controls such as human or mouse cell lines expressing FUT8.
- Polyclonal nature may provide robust signal in ELISA; optimize coating and detection conditions for your assay format.
