Biological Background: FUT7 Function and Localization

  • FUT7 (Fucosyltransferase VII) is a Golgi-resident glycosyltransferase that catalyzes the transfer of L-fucose to sialylated N-acetyllactosamine units, a key step in the synthesis of sialyl Lewis X (sLe(x)) epitopes (PubMed:11404359, PubMed:15632313, PubMed:15926890). Related references: PMID:11404359 PMID:15632313 PMID:15926890
  • Through sLe(x) biosynthesis, FUT7 controls selectin-mediated leukocyte adhesion, enabling tethering and rolling along endothelial tissue during inflammation (PubMed:10386892, PubMed:29138114, PubMed:8666674). Related references: PMID:10386892 PMID:29138114 PMID:8666674
  • In vitro, FUT7 can synthesize sialyl-dimeric-Lex and multi-fucosylated structures, but its activity is inhibited by internal alpha1-3 fucosylation on polylactosamine chains (PubMed:9499379, PubMed:9473504). Related references: PMID:9499379 PMID:9473504
  • FUT7 transfers fucose to 6-sulfated sialylated substrates to produce 6-sulfo sLex, mediating L-selectin-dependent cell adhesion (PubMed:10200296, PubMed:8752218). Related references: PMID:10200296 PMID:8752218
  • The enzyme localizes specifically to the Golgi apparatus membrane.
  • Expression is restricted to leukocytic/myeloid lineage cells.
  • FUT7 may enhance embryo implantation via sLe(x)-mediated embryo-endometrium adhesion and modulate insulin signaling by modifying INSRR glycans (PubMed:18402946, PubMed:18553500, PubMed:17229154). Related references: PMID:18402946 PMID:18553500 PMID:17229154

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 100-200 of human FUT7. Antigenic regions may influence detection specificity.
  • For WB, a band around 39 kDa is expected. Include appropriate controls to confirm specificity, especially across human, mouse, and rat samples.
  • For ELISA, optimize coating antigen and antibody dilution to achieve optimal signal-to-noise ratio.
  • Since FUT7 is Golgi-localized, consider cellular fractionation or specific lysate preparation to enrich for membrane proteins.
