Biological Background: FUT6 Function and Localization
- FUT6 (Fucosyltransferase 6), also designated Fuc-TVI, is a glycosyltransferase belonging to the alpha-1,3-fucosyltransferase family. It catalyzes the transfer of L-fucose from GDP-beta-L-fucose to the N-acetylglucosamine (GlcNAc) of acceptor substrates, participating in the biosynthesis of sialyl Lewis X (sLe(x)), Lewis X (Le(x)), and VIM2 determinants. - FUT6 exhibits specificity for distal alpha2,3-sialylated and neutral polylactosamine chains on glycoproteins and glycolipids. It can further fucosylate H-type 2 chain acceptors to generate difucosylated sialyl Lewis x structures but lacks activity toward type 1 and H-type 1 chain substrates. - At the subcellular level, FUT6 localizes primarily to the Golgi stack membrane, where it functions as a resident type II membrane protein, and is also found in secreted forms.

- The immunogen used to generate this antibody encompasses amino acids 170–359 of human FUT6, a region covering the catalytic domain. This design likely enables recognition of both membrane-bound and secreted forms of the enzyme.
- In Western blotting (WB), the predicted molecular weight is approximately 42 kDa; however, glycosylation may cause slight shifts. Validate antibody binding under reducing and non-reducing conditions in your relevant sample system.
- For immunohistochemistry (IHC-P), consider optimal antigen retrieval methods and include negative controls to distinguish specific staining, especially given the Golgi localization pattern.
- ELISA applications can benefit from titrating the antibody against the recombinant immunogen or native FUT6 to determine appropriate working concentrations.
