Biological Background: FUT6 Function and Localization

  • FUT6 (Fucosyltransferase 6), also designated Fuc-TVI, is a glycosyltransferase belonging to the alpha-1,3-fucosyltransferase family. It catalyzes the transfer of L-fucose from GDP-beta-L-fucose to the N-acetylglucosamine (GlcNAc) of acceptor substrates, participating in the biosynthesis of sialyl Lewis X (sLe(x)), Lewis X (Le(x)), and VIM2 determinants.   • FUT6 exhibits specificity for distal alpha2,3-sialylated and neutral polylactosamine chains on glycoproteins and glycolipids. It can further fucosylate H-type 2 chain acceptors to generate difucosylated sialyl Lewis x structures but lacks activity toward type 1 and H-type 1 chain substrates.
