Biological Background: FUT4 Function and Localization
- Encodes alpha-(1,3)-fucosyltransferase 4 (FUT4), a Golgi-resident transmembrane enzyme.
- Catalyzes the transfer of fucose from GDP-Beta-L-fucose to N-acetylglucosamine (GlcNAc) within type 2 lactosamine (LacNAc) to form the Lewis X antigen (CD15) (PubMed:1702034, PubMed:1716630, PubMed:29593094). Related references: PMID:1702034 PMID:1716630 PMID:29593094
- Also fucosylates sialylated LacNAc acceptors, generating sialyl Lewis X and 6-sulfo sialyl Lewis X determinants that serve as C-type lectin epitopes (PubMed:1716630, PubMed:29593094). Related references: PMID:1716630 PMID:29593094
- Cooperates with FUT7 to synthesize selectin ligands (e.g., SELE, SELL, SELP), critical for lymphocyte homing, leukocyte migration, and blood leukocyte homeostasis (By similarity).
- In a cell-type-specific context, can fucosylate internal LacNAc units of polylactosamine chains to produce VIM-2 antigen, a recognition determinant for SELE (PubMed:11278338, PubMed:1716630). Related references: PMID:11278338 PMID:1716630
- Localized to the Golgi stack membrane.
- Expressed in bone marrow-derived mesenchymal stem cells, cord blood immature promyelocytes, and peripheral blood myeloid and lymphoid populations.
- Classified under keywords: Alternative initiation, Glycoprotein, Glycosyltransferase, Golgi apparatus, Inflammatory response, Membrane, Signal-anchor, Transferase, Transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen corresponds to an internal region (aa 186-331) of human FUT4; thus the antibody detects an epitope likely conserved across species (cross-reacts with mouse and rat).
- For Western blotting, electrophorese samples under denaturing conditions and probe for a band at ~59 kDa. Consider using HL-60 or U937 cell lysates as positive controls.
- For immunofluorescence and immunohistochemistry, anticipate a Golgi-like perinuclear staining pattern. Use appropriate fixation and permeabilization conditions for organelle visualization.
- ELISA protocols may be optimized using recombinant FUT4 protein or peptide fragments as coating antigens.
- Validate antibody performance in your specific sample system and assay conditions before drawing conclusions.
