Biological Background: FUT2 Function and Localization
- FUT2 encodes galactoside alpha-(1,2)-fucosyltransferase 2, also known as secretor factor (Se) and fucosyltransferase 2.
- Catalyzes the transfer of L-fucose to terminal galactose on glycans, creating the H antigen essential for ABO blood group synthesis. Related references: PMID:7876235
- Preferentially fucosylates gangliosides GA1 and GM1 in the gastrointestinal and reproductive tissues, mediating host-microbe interactions. Related references: PMID:21625510, PMID:22068912, PMID:24733310
- Localizes to the Golgi apparatus, Golgi stack membrane.
- Expressed in small intestine, colon, and lung.
- Keywords: Blood group antigen, Glycoprotein, Glycosyltransferase, Golgi apparatus, Membrane, Transferase, Transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 29–150 of human FUT2, covering the N-terminal region. This design may facilitate recognition of both glycosylated and unglycosylated forms.
- For Western blot (WB), the predicted molecular weight is approximately 39 kDa. Consider running appropriate molecular weight markers and validating expression in relevant tissues (e.g., small intestine, colon, lung) where FUT2 is known to be expressed.
- For ELISA, optimization of coating antigen and antibody concentration is recommended. Use proper controls to verify specificity.
- Note that FUT2 is a glycosyltransferase with transmembrane domain; sample preparation should consider membrane protein extraction protocols if targeting the endogenous protein.
