Biological Background: FUS Function and Localization

  • RNA-binding protein FUS (also designated TLS) is a multifunctional DNA/RNA-binding protein involved in transcription regulation, RNA splicing, RNA transport, and DNA damage repair. Related references: PMID:27731383
  • Binds specifically to ssRNA with the consensus sequence 5'-AGGUAA-3' and to nascent pre-mRNAs, mediating the coupling of transcription and splicing via RNA polymerase II and U1 snRNP. Related references: PMID:21256132, PMID:26124092
  • Autoregulates its own expression through binding to its pre-mRNA, a mechanism mediated by nonsense-mediated decay. Related references: PMID:24204307
  • Facilitates DNA double-strand break repair by promoting D-loop formation and homologous recombination. Related references: PMID:10567410
  • In neuronal cells, plays critical roles in dendritic spine formation and stability, RNA transport, mRNA stability, and synaptic homeostasis.
  • Subcellular localization is predominantly nuclear.
  • Ubiquitously expressed; the protein features zinc finger and RNA recognition motifs, undergoes post-translational modifications including phosphorylation, methylation, and ubiquitination, and is associated with amyotrophic lateral sclerosis and oncogenic rearrangements.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 297–526 within the C-terminal region of human FUS, which includes the zinc finger domain and a portion of the RNA recognition motifs.
  • This rabbit polyclonal antibody detects endogenous FUS in multiple applications: Western blot (expected band ~53 kDa), immunohistochemistry on paraffin-embedded sections, immunoprecipitation, and ELISA.
  • Reported cross-reactivity with mouse and rat FUS; however, due to potential species-specific variations, validation in the target sample system is recommended.
  • For IHC-P, consider antigen retrieval optimization and positive control tissues such as brain or testis.
  • As FUS is subject to post-translational modifications, additional bands may appear in some cell lines; verify band patterns with appropriate controls.
