Biological Background: Furin Function and Localization
- Furin, also known as PACE, PCSK3, or SPC1, is a member of the proprotein convertase family encoded by the FURIN gene.
- Functions as a ubiquitous endoprotease within constitutive secretory pathways, cleaving at the RX(K/R)R consensus motif (PubMed:11799113, PubMed:1629222, PubMed:1713771). Related references: PMID:11799113, PMID:1629222, PMID:1713771
- Mediates proteolytic processing of TGFB1, an essential step for TGF-beta-1 activation (PubMed:7737999). Related references: PMID:7737999

Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 200-300 of human Furin; consider that the antibody may preferentially recognize epitopes in this region.
- For Western blot, an expected band around 87 kDa is observed, but due to zymogen processing and autocatalytic cleavage, additional bands corresponding to pro- and mature forms may appear. Validate band patterns in appropriate cell lines or tissues.
- For immunofluorescence/ICC, optimized fixation and permeabilization conditions may be required to detect Golgi and TGN localization. Include appropriate controls.
- This polyclonal antibody is suitable for ELISA; recommend titrating to determine optimal working dilutions for specific assay conditions.
- The antibody cross-reacts with mouse and rat Furin, allowing use in those species; always validate in the relevant sample system.
