Biological Background: Furin Function and Localization
- Furin (PCSK3) is a calcium-dependent serine endoprotease belonging to the proprotein convertase family, also known as paired basic amino acid residue-cleaving enzyme (PACE) and dibasic-processing enzyme.
- Furin recognizes and cleaves polypeptide precursors at the consensus sequence RX(K/R)R, playing a critical role in proprotein maturation within the constitutive secretory pathway. Related references: PMID:11799113, PMID:1629222, PMID:1713771
- It processes key proteins such as transforming growth factor beta-1 (TGFB1) and brain natriuretic factor prohormone, converting them into their active forms. Related references: PMID:7737999, PMID:20489134, PMID:21763278
- Furin cycles between the trans-Golgi network, cell surface, and endosomes, and is also secreted in a soluble form.

- Furin is ubiquitously expressed in human tissues, reflecting its central role in processing a wide array of secreted and membrane proteins.

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from amino acids 200–300; the antibody likely recognizes a linear epitope, making it suitable for denaturing applications such as Western blot. For native applications like ELISA, validate binding under non-denaturing conditions.
- In Western blot, expect a band around 87 kDa for full-length Furin. Due to proprotein processing, additional bands corresponding to the proform and mature cleaved forms may also appear.
- Ubiquitous expression allows many cell lines to serve as positive controls; consider HeLa, HEK293, or A549 cells. Cross-reactivity with mouse is indicated, but verify sequence homology in the immunogen region when testing other species.
- For ELISA, optimize coating conditions and antibody titers, as the peptide immunogen may influence recognition of the native protein.
