Biological Background: FTO Function and Localization
- FTO (Fat mass and obesity-associated protein) is an alpha-ketoglutarate-dependent dioxygenase that functions as an RNA demethylase. Also known as ALKBH9, it belongs to the AlkB family of dioxygenases.
- It mediates oxidative demethylation of N6-methyladenosine (m6A) in mRNA, the most prevalent internal modification in higher eukaryotes, thereby regulating mRNA expression and stability. Related references: PMID:22002720, PMID:25452335, PMID:26457839, PMID:30197295

Experimental Guidance and Technical Tips
- The immunogen is a peptide from the N-terminal region (aa 1-100), which may detect full-length FTO and possibly some isoforms. Include positive and negative controls (e.g., FTO-overexpressing lysates or siRNA-treated samples) to confirm specificity.
- For Western blot (WB), run reduced and denatured protein samples; the expected band size is ~58 kDa. Use appropriate loading controls and blocking conditions.
- In IHC-P and IF/ICC, test different fixation and antigen retrieval methods. Paraffin-embedded sections may require heat-induced epitope retrieval. Validate staining using known positive tissues (brain, adrenal) and include isotype controls.
- For immunoprecipitation (IP), the antibody may be suitable for capturing native FTO complexes. Optimize lysis buffer conditions and antibody amount per given total protein.
- For ELISA, the antibody can be utilized in both direct and sandwich formats; optimize coating and detection conditions according to your specific assay requirements.
