Biological Background: FTO Function and Localization

  • FTO (Fat mass and obesity-associated protein) is an alpha-ketoglutarate-dependent dioxygenase that functions as an RNA demethylase. Also known as ALKBH9, it belongs to the AlkB family of dioxygenases.

Experimental Guidance and Technical Tips

  • For Western blot (WB), run reduced and denatured protein samples; the expected band size is ~58 kDa. Use appropriate loading controls and blocking conditions.
  • In IHC-P and IF/ICC, test different fixation and antigen retrieval methods. Paraffin-embedded sections may require heat-induced epitope retrieval. Validate staining using known positive tissues (brain, adrenal) and include isotype controls.
