Biological Background: S100A4 Function and Localization

  • S100A4, also known as FSP1, calvasculin, metastasin, Mts1, CAPL, and p9Ka, is a member of the S100 family of calcium-binding proteins containing EF-hand motifs.
  • Plays a central role in cell motility and metastasis by interacting with non-muscle myosin heavy chain IIA (MYH9), promoting filament depolymerization and increasing soluble myosin-IIA to facilitate chemotaxis. Related references: PMID:16707441
  • Modulates apoptosis by binding the C-terminal transactivation domain of TP53 within the nucleus, leading to reduced TP53 protein levels and inhibition of its pro-apoptotic function. Related references: PMID:23752197
  • Secreted extracellularly, S100A4 acts as a cytokine, stimulating granulocyte colony-stimulating factor and CCL24 production from T-lymphocytes, and functions as a chemoattractant when complexed with PGLYRP1, promoting lymphocyte migration via CCR5 and CXCR3 receptors. Related references: PMID:30713770
  • Subcellular localization includes cytoplasm, nucleus, and secretion, consistent with its multifunctional roles.
  • Ubiquitously expressed in human tissues.
  • Keywords: calcium-binding, metal-binding, acetylation, and involvement in cell differentiation and autophagy.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the nearly full-length S100A4 protein (amino acids 2-101), excluding the initiator methionine, and may recognize both native and denatured forms.
  • For Western blot, consider using whole-cell lysates from HeLa or A431 cells known to express S100A4. Due to the low molecular weight (12 kDa), ensure the blotting membrane has an appropriate pore size (0.2 µm or smaller) and optimize transfer conditions.
  • In immunofluorescence and immunohistochemistry, permeabilization is recommended to detect nuclear and cytoplasmic pools. Validate in the relevant sample system.
  • Cross-reactivity with mouse and rat suggests a conserved epitope; confirm performance in the species of interest.
