Biological Background: FRS2 Function and Localization

  • FRS2 (Fibroblast growth factor receptor substrate 2) is an adaptor protein that links activated FGF and NGF receptors to downstream signaling cascades, including the MAP kinase pathway and PI3K/AKT pathway via phosphorylation of PIK3R1.
  • It modulates signaling through SHC1 by competing for a common binding site on NTRK1, thereby influencing neurotrophin signaling.
  • The protein localizes to the endomembrane system.
  • FRS2 is highly expressed in heart, brain, spleen, lung, liver, skeletal muscle, kidney, and testis.
  • Post-translational modifications include myristoylation and phosphorylation; it is also subject to ubiquitination (Ubl conjugation).
