Biological Background: Frataxin Function and Localization

Frataxin (FXN) is a mitochondrial protein that functions as a critical activator of persulfide transfer to the scaffold protein ISCU within the core iron-sulfur cluster (ISC) assembly complex, essential for de novo [2Fe-2S] cluster biogenesis.

The core ISC complex, consisting of NFS1:LYRM4:NDUFAB1 and ISCU, produces persulfide that is delivered to ISCU in an FXN-dependent manner; the complex is then stabilized by FDX2, enabling [2Fe-2S] cluster assembly and transfer to chaperones (HSCB, HSPA9, GLRX5) (By similarity).

FXN binds ferrous ion (Fe²⁺) and is released upon addition of L-cysteine and reduced FDX2 during cluster assembly; it accelerates sulfur transfer from NFS1 persulfide intermediate to ISCU and small thiols like L-cysteine and glutathione, promoting persulfuration and sulfide release.
