Biological Background: Frataxin Function and Localization
- Encoded by the FXN gene (also known as FRDA or X25), frataxin is a 23-kDa mitochondrial protein that is highly conserved. Expansions of an intronic GAA triplet repeat cause Friedreich ataxia, a neurodegenerative disease.
- Frataxin is a key component of the core iron-sulfur cluster (ISC) assembly complex in mitochondria, where it activates persulfide transfer from the cysteine desulfurase complex NFS1:LYRM4:NDUFAB1 to the scaffold protein ISCU, enabling de novo [2Fe-2S] cluster synthesis. - It binds ferrous iron and is released upon addition of L-cysteine and reduced FDX2, facilitating [2Fe-2S] cluster assembly. The mature cluster is then transferred to chaperones like HSCB, HSPA9, and GLRX5. Related references: PMID:29576242
