Biological Background: FOXO3A Function and Localization
- FOXO3A (Forkhead box protein O3) is encoded by the Foxo3 gene (also known as Fkhr2, Foxo3a) in mouse and belongs to the forkhead family of transcription factors.
- As a transcriptional activator, it specifically binds the DNA motif 5'-[AG]TAAA[TC]A-3' and regulates key processes including apoptosis and autophagy. Related references: PMID:18054315 PMID:18054316 PMID:23805378
- In skeletal muscle, FOXO3A acts as a positive autophagy regulator: upon starvation, dephosphorylated FOXO3A enters the nucleus, binds autophagy gene promoters (GABARAP1L, MAP1LC3B, ATG12), and activates their expression, causing proteolysis. - FOXO3A triggers apoptosis in the absence of survival factors, including neuronal cell death upon oxidative stress (by similarity), and participates in post-transcriptional MYC regulation by promoting miR-34b/c expression after MAPKAPK5 phosphorylation (by similarity).
- Under metabolic stress, FOXO3A translocates into mitochondria and enhances mtDNA transcription; when lipid levels are low, nuclear FOXO3A induces SOX9 expression to drive chondrogenic commitment and suppress fatty acid oxidation. Related references: PMID:23283301 PMID:32103177
- The protein localizes to cytoplasm, cytosol, nucleus, and mitochondrial matrix/outer membrane, and is expressed in white/brown adipose tissue, liver, kidney, lung, colon, and skeletal muscle. Post-translational modifications include phosphorylation, acetylation, ubiquitination, and methylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 558–635 near the C-terminus of mouse FOXO3A; given the high conservation across human and rat, this antibody is expected to cross-react with these species, as indicated.
- In Western blot, a band around 71 kDa is anticipated; validate molecular weight and specificity in your sample system.
- For IHC-P and IF/ICC, optimize fixation, antigen retrieval, and permeabilization protocols to achieve clear staining.
- The antibody is also suitable for ELISA; include appropriate positive and negative controls in all assays.
