Biological Background: FOXO3A Function and Localization

  • FOXO3A (Forkhead box protein O3) is encoded by the Foxo3 gene (also known as Fkhr2, Foxo3a) in mouse and belongs to the forkhead family of transcription factors.
  • As a transcriptional activator, it specifically binds the DNA motif 5'-[AG]TAAA[TC]A-3' and regulates key processes including apoptosis and autophagy. Related references: PMID:18054315 PMID:18054316 PMID:23805378
  • In skeletal muscle, FOXO3A acts as a positive autophagy regulator: upon starvation, dephosphorylated FOXO3A enters the nucleus, binds autophagy gene promoters (GABARAP1L, MAP1LC3B, ATG12), and activates their expression, causing proteolysis.   • FOXO3A triggers apoptosis in the absence of survival factors, including neuronal cell death upon oxidative stress (by similarity), and participates in post-transcriptional MYC regulation by promoting miR-34b/c expression after MAPKAPK5 phosphorylation (by similarity).
