Biological Background: FOXO3A Function and Localization
- FOXO3A is a forkhead box O family transcriptional activator that binds DNA motif 5'-[AG]TAAA[TC]A-3', regulating apoptosis and autophagy. Related references: PMID:10102273 PMID:16751106 PMID:21329882
- In starved conditions, it translocates to the nucleus and induces autophagy gene expression (e.g., GABARAP1L, MAP1LC3B, ATG12), promoting muscle protein breakdown.
- Phosphorylation by MAPKAPK5 enables FOXO3A to upregulate miR-34b/c, which in turn repress MYC translation. Related references: PMID:21329882
- Upon metabolic stress, FOXO3A localizes to mitochondria and enhances mtDNA transcription. Related references: PMID:23283301
- It is ubiquitously expressed and shuttles between cytoplasm and nucleus, with additional mitochondrial matrix and outer membrane localization.
- FOXO3A is a key regulator of Treg differentiation via FOXP3 activation. Related references: PMID:30513302
- Post-translational modifications—including phosphorylation, acetylation, methylation, and ubiquitination—modulate its activity, stability, and subcellular distribution.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (aa 574–673), which may target specific isoforms. Verify reactivity with your sample.
- For WB, the predicted molecular weight is ~71 kDa; consider appropriate controls to confirm specificity, especially given potential post-translational modifications.
- ELISA can be used for quantitative detection; optimize coating and detection conditions for your assay format.
- Since FOXO3A exhibits multiple subcellular localizations, fractionation or immunofluorescence may be necessary to study context-dependent translocation.
