Biological Background: FOXO3A Function and Localization

  • Forkhead box protein O3 (FOXO3A), also known as Fkhr2 or Foxo3a, is encoded by the Foxo3 gene.
  • FOXO3A is a transcriptional activator that binds the consensus DNA sequence 5'-[AG]TAAA[TC]A-3', regulating apoptosis and autophagy (PubMed:18054315, PubMed:18054316, PubMed:23805378). Related references: PMID:18054315 PMID:18054316 PMID:23805378
  • It positively regulates autophagy in skeletal muscle by dephosphorylation, nuclear entry, and activation of autophagy genes like GABARAP1L and MAP1LC3B, leading to proteolysis (PubMed:18054315, PubMed:18054316, PubMed:25402684). Related references: PMID:18054315 PMID:18054316 PMID:25402684
  • In response to metabolic stress, FOXO3A translocates to mitochondria to promote mtDNA transcription (PubMed:23283301). Related references: PMID:23283301
  • The protein shuttles between the cytoplasm, nucleus, and mitochondria (matrix and outer membrane), with localization changes depending on cellular conditions.
  • Tissue expression includes white and brown adipose tissues, liver, kidney, lung, colon, and skeletal muscle (PubMed:22510882, PubMed:29445193). Related references: PMID:22510882 PMID:29445193
  • Post-translational modifications include phosphorylation, acetylation, methylation, and ubiquitination, which regulate its activity and stability.
  • Additionally, FOXO3A participates in post-transcriptional regulation of MYC via miR-34b/c, promotes chondrogenic commitment of skeletal progenitors (PubMed:32103177), and drives regulatory T-cell differentiation by activating FOXP3. Related references: PMID:32103177

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 371–470 of mouse FOXO3A; the resulting polyclonal antibodies may recognize multiple epitopes, making them well-suited for denaturing Western blotting where linearized epitopes are exposed.
  • For WB, expect a band at approximately 71 kDa; however, phosphorylation and other modifications may shift or create additional bands, so validate with appropriate controls and phosphatase inhibitors if needed.
  • For IF/ICC and IF-P, optimize fixation and permeabilization conditions to ensure antibody access to nuclear and cytoplasmic compartments where FOXO3A localizes.
  • For ELISA, this antibody can be considered as either a capture or detection reagent, but users should perform checkerboard titrations to determine optimal working concentrations.
  • Cross-reactivity has been confirmed with mouse and rat; reactions with other species should be experimentally verified.
