Biological Background: FOXM1 Function and Localization
- FOXM1 is a transcription factor that regulates the expression of cell cycle genes essential for DNA replication and mitosis, playing a key role in cell proliferation. Related references: PMID:19160488, PMID:20360045
- It is involved in DNA break repair and the DNA damage checkpoint response, and promotes transcription of prohibitin-2 (PHB2). Related references: PMID:17101782, PMID:33754036
- FOXM1 promotes epithelial-mesenchymal transition (EMT) by activating Rho-associated protein kinases ROCK1 and ROCK2. Related references: PMID:27399334
- It enhances PI3K/AKT signaling by binding to both catalytic and regulatory subunits of PI3K, leading to AKT activation and increased SNAI1 levels. Related references: PMID:32606397
- FOXM1 inhibits glycolysis by binding to the pyruvate kinase PKM homotetramer, and promotes nuclear translocation of PKM and NF-κB to enhance VEGFA transcription and angiogenesis. Related references: PMID:33314660
- The protein localizes to both the nucleus and cytoplasm.
- It is predominantly expressed in proliferating cells and tissues such as thymus, testis, and colon, and is not expressed in resting cells. Isoform 2 is highly expressed in testis.
- Keywords include phosphoprotein, DNA-binding, cell cycle, transcription regulation, and ubiquitin conjugation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 29–128 of human FOXM1 (N-terminal region). Consider this when designing experiments or validating in the relevant sample system.
- For Western blot (WB), the predicted molecular weight is 84 kDa, but multiple isoforms may be detected. Validate in your sample type.
- This antibody has been shown to detect FOXM1 in human, mouse, and rat samples; cross-reactivity with other species should be tested individually.
- Suitable applications include WB, immunofluorescence (IF-P), and ELISA. Optimization of dilution and conditions is recommended for each assay.
