Biological Background: FOXK2 Function and Localization

  • FOXK2 encodes a forkhead box transcription factor with multiple isoforms produced by alternative splicing; also known as ILF, ILF1, and nGTBP.
  • Recognizes the forkhead motif 5'-GTAAACA-3' and acts as a context-dependent transcriptional activator or repressor (PubMed:22083952, PubMed:25451922). Additionally binds NFAT-like motifs in the IL2 promoter (PubMed:1339390) and the HIV-1 long terminal repeat (PubMed:1909027).   • Plays a key role in metabolic reprogramming toward aerobic glycolysis by inducing glycolytic enzymes (HK2, PKLR, LDHA) and PDK1/4, while suppressing mitochondrial oxidation (By similarity).
