Biological Background: FOXK1 Function and Localization
- FOXK1 (Forkhead box protein K1, also known as MNF) is a transcription factor that binds the forkhead DNA motif (5'-GTAAACA-3') and can act as an activator or repressor.
Related references: PMID:17670796 - It regulates aerobic glycolysis by inducing glycolytic enzymes (HK2, PKLR) and suppressing mitochondrial pyruvate oxidation via PDK1/PDK4.
- FOXK1 promotes muscle progenitor cell proliferation and inhibits myogenic differentiation by repressing FOXO4 and MEFC activity.
- It positively regulates Wnt/β-catenin signaling by translocating DVL into the nucleus.
Related references: PMID:25805136 - Exhibits antiviral activity by binding ISRE to promote antiviral gene expression.
Related references: PMID:25852164 - Acts as an accessory component of the PR-DUB deubiquitinase complex, recruiting it to specific genes.
Related references: PMID:24634419 PMID:30664650 - Phosphorylation by PRKCB isoform β-II promotes ferroptosis via LGALS13 expression and secretion.
Related references: PMID:40246981 - Subcellular location: primarily nuclear, also cytoplasmic. Expressed in developing and adult tissues, with significant expression in brain, colon, and lymph node tumors. Numerous post-translational modifications include acetylation, methylation, and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen covers the C-terminal region (aa 600-733) of human FOXK1; the antibody may recognize both full-length and potential splice variants.
- For Western blotting, the predicted molecular weight is ~75 kDa. Validate with positive and negative cell lysates, and consider using loading controls.
- For ELISA, appropriate antigen coating and blocking conditions should be optimized; include known positive and negative controls.
- Since cross-reactivity is reported for human, perform species-specific validation if used in other species.
