Biological Background: FOXI1 Function and Localization
- FOXI1, also known as Forkhead box protein I1, FKHL10, FREAC6, or HFH3, is a member of the forkhead/winged helix transcription factor family characterized by a conserved DNA-binding domain.
- Acts as a transcriptional activator essential for the development of normal hearing, sense of balance, and kidney function by regulating the expression of key target genes such as SLC26A4/PDS, JAG1, and COCH in inner ear epithelial cells, and SLC4A1/AE1, SLC4A9/AE4, ATP6V1B1 in renal intercalated cells (By similarity).
- Localizes to the nucleus where it exerts its transcriptional regulatory functions.
- Expressed specifically in the kidney.
- Undergoes alternative splicing, producing multiple isoforms that may have distinct functional roles.
- Required for the differentiation of intercalated cells in the epithelium of distal renal tubules, highlighting its role in epithelial cell specialization.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human FOXI1; for Western blot detection, consider using lysates from kidney or inner ear tissues where FOXI1 is expressed.
- For ELISA applications, the peptide immunogen can serve as a positive control; validate binding under your specific assay conditions.
- Due to cross-reactivity with mouse and rat, this antibody may be suitable for comparative studies; however, verify species-specific reactivity in your experimental system.
