Biological Background: FOXC1 Function and Localization
- FOXC1 (Forkhead box protein C1) is a member of the forkhead family of transcription factors, also known as FKHL7 and FREAC-3, characterized by a conserved forkhead DNA-binding domain.
- FOXC1 functions as a DNA-binding transcriptional activator or repressor, recognizing the consensus sequence 5'-[G/C][A/T]AAA[T/C]AA[A/C]-3' and inducing DNA bending upon binding. Related references: PMID:11782474 PMID:12533514 PMID:14506133 PMID:7957066
- It plays critical roles in embryonic development, including eye, bone, cardiovascular, kidney, and skin formation, and acts as a transcriptional coactivator for GLI2 in hedgehog signaling during endochondral ossification. Related references: PMID:11782474 PMID:14578375 PMID:26565916
- FOXC1 regulates FOXO1 expression through promoter binding, thereby influencing cell viability and resistance to oxidative stress, particularly in ocular tissues. Related references: PMID:17993506
- It is involved in cell cycle control by promoting G1 phase arrest via TGFB1-mediated signals, suggesting tumor suppressor activity. Related references: PMID:12408963
- Additionally, FOXC1 promotes epithelial-mesenchymal transition (EMT), cell proliferation, migration, and invasion, and is highly expressed in basal-like breast cancer and hepatocellular tumors. Related references: PMID:20406990 PMID:22991501
- FOXC1 is essential for epidermal keratinocyte terminal differentiation and is expressed in the nucleus of keratinocytes, microvascular invasion formations, and hematopoietic cells. Related references: PMID:27907090 PMID:8499623
Experimental Guidance and Technical Tips
- The immunogen covers the C-terminal region (aa 404-553) of human FOXC1, which may be useful for detecting both full-length and potential C-terminal variants; however, users should verify with appropriate controls.
- For Western blot, the predicted molecular weight is approximately 57 kDa; consider optimizing primary antibody dilution (e.g., 1:500-1:2000) and using lysates from cells known to express FOXC1 (e.g., breast cancer, keratinocyte lines).
- For ELISA, ensure the coating antigen includes or overlaps with the immunogen region to maximize detection sensitivity.
- Cross-reactivity with mouse and rat is predicted based on sequence similarity; validate in the relevant species model if needed.
