Biological Background: FOXA1 Function and Localization

  • FOXA1 (Forkhead box protein A1; HNF3A; TCF3A) is a pioneer transcription factor that regulates gene expression by opening chromatin and recruiting other transcription factors to enhancer/promoter regions.
  • Binds DNA via a conserved forkhead domain recognizing the consensus 5'-[AC]A[AT]T[AG]TT[GT][AG][CT]T[CT]-3' and facilitates cell-type-specific transcriptional programs dependent on histone modifications such as H3K4me2.
  • Essential for embryonic development of endoderm-derived organs including the liver, lung, prostate, and pancreas; cooperates with FOXA2 in these processes.
  • Modulates nuclear hormone receptor signaling, critically required for ESR1 (estrogen receptor) binding to target promoters in breast cancer and for estrogen-dependent gene regulation.
  • Controls cell survival and proliferation by repressing BCL2 (anti-apoptotic) and activating CDKN1B (p27/Kip1 cell cycle inhibitor), partly through interaction with BRCA1.
  • Activates transcription of liver-specific genes (albumin, AFP, PEPCK) and contributes to metabolic homeostasis including glucose regulation.
  • Found predominantly in the nucleus; highly expressed in prostate, ESR1-positive breast tumors, and overexpressed in lung and esophageal adenocarcinomas.
  • Undergoes alternative splicing and phosphorylation, consistent with its regulation at multiple levels.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 263-472) of human FOXA1, which includes part of the forkhead domain and transactivation domain; this region is highly conserved across human, mouse, and rat, supporting cross-reactivity.
  • For WB, a band near 49 kDa is expected; consider using nuclear extracts from FOXA1-expressing tissues or cell lines (e.g., liver, prostate, breast cancer) and validate antibody specificity with appropriate controls.
  • IHC-P may require antigen retrieval and optimization of antibody dilution; nuclear staining is anticipated. Validate on positive control tissues such as human liver or prostate adenocarcinoma.
  • For ELISA, the antibody can be tested as a capture or detection reagent; pairing with a monoclonal anti-FOXA1 antibody may improve sandwich assay performance after in-house validation.
