Biological Background: FOSB Function and Localization

  • FOSB encodes the protein FosB, a member of the AP-1 transcription factor complex subunit family, also referred to as G0S3, GOS3, GOSB, and FOSB.
  • Heterodimerizes with JUN family proteins to form the AP-1 transcription factor complex, enhancing DNA binding to the AP-1 consensus sequence 5'-TGA[GC]TCA-3' and promoting transcriptional activity. Related references: PMID:12618758 PMID:28981703
  • As part of the AP-1 complex, facilitates enhancer selection by cooperatively binding to nucleosomal enhancers and recruiting the SWI/SNF (BAF) chromatin remodeling complex to establish accessible chromatin (By similarity).
  • Together with JUN, participates in activation-induced cell death of T cells by binding to the AP-1 promoter site of FASLG/CD95L and inducing its transcription in response to TCR/CD3 signaling. Related references: PMID:12618758
  • Alternative splicing produces isoforms; isoform 2 shows lower transactivation activity than isoform 1 and forms an inactive heterodimer with JUN, potentially acting as a transcriptional inhibitor (By similarity).

  • This rabbit polyclonal antibody was raised against an N-terminal synthetic peptide (amino acids 1-100) of human FOSB; sequence homology with mouse and rat FOSB may account for the observed cross-reactivity.
  • In western blot, a band near 36 kDa is expected; however, additional bands or shifts may occur due to post-translational modifications or isoform expression, warranting validation in the relevant sample system.
  • For immunohistochemistry (paraffin-embedded sections), nuclear staining should be observed based on FOSB localization; optimize antigen retrieval and primary antibody dilution for each tissue type.
  • In ELISA, consider employing the antibody as either a capture or detection reagent, with appropriate controls to ensure specificity.
