Biological Background: c-Fos Function and Localization

  • Encodes a nuclear phosphoprotein that forms a tight non-covalent heterodimer with JUN/AP-1 transcription factor, binding to symmetrical DNA half sites to regulate gene expression.
  • On TGF-beta activation, assembles into a multimeric SMAD3/SMAD4/JUN/FOS complex at AP1/SMAD-binding sites, mediating TGF-beta signaling.
  • Plays a critical role in skeletal development, maintaining cells destined to form and maintain the skeleton.
  • Involved in signal transduction, cell proliferation, and differentiation.
  • In growing cells, activates phospholipid synthesis by possibly activating CDS1 and PI4K2A, requiring Tyr-dephosphorylation and association with the endoplasmic reticulum.
  • Localizes to the nucleus, endoplasmic reticulum, cytoplasm, and cytosol.
  • Identified as a proto-oncogene; subjected to alternative splicing, phosphorylation, and ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of human FOS, suggesting the antibody may detect multiple isoforms expressed through alternative splicing.
  • For western blot, consider using whole cell lysates from serum-stimulated or growth factor-treated cell lines to upregulate FOS expression; the predicted molecular weight is approximately 41 kDa.
  • In ELISA, the peptide-based immunogen may enable detection of c-Fos in denatured or native lysates; include appropriate peptide or recombinant standards for quantification.
  • Cross-reactivity with mouse and rat indicates conserved epitope; however, validate in the desired species before large-scale experiments.
