Biological Background: Follistatin Function and Localization

  • Follistatin is a secreted regulatory protein that binds and neutralizes TGF-beta superfamily ligands, including activin, myostatin, GDF11, and BMPs, by obstructing their type II receptor-binding site and inhibiting downstream signaling cascades.   • As a specific inhibitor of pituitary follicle-stimulating hormone (FSH), follistatin sequesters activin A, reducing FSH biosynthesis and secretion. Related references: PMID:11279126
  • It plays an essential role in muscle fiber formation and growth by counteracting myostatin and also through SMAD3/AKT/mTOR signaling independently of myostatin.
  • Follistatin promotes neural differentiation by antagonizing BMP4 action.
