Biological Background: FNDC4 Function and Localization
- Fibronectin type III domain-containing protein 4 (FNDC4), also known as fibronectin type III repeat-containing protein 1 (FRCP1), is a member of the fibronectin type III domain-containing protein family.
- Has anti-inflammatory properties; in the colon, it acts on macrophages to down-regulate inflammation.
- May suppress osteoclastogenesis and mature osteoclast resorptive function, suggesting a role in bone metabolism.
- In white adipose tissue, decreases local inflammation via interaction with GPR116, and is required for proper systemic glucose tolerance by sensitizing white adipocytes to insulin and promoting glucose uptake.
- The insulin-sensitizing function in adipose tissue is mediated through interaction with ADGRF5/GPR116 and activation of cAMP signaling.
- Localizes to both the membrane and secreted compartments, consistent with its role as a signaling molecule.
- Highly expressed in liver and brain, including the cortex, hypothalamus, and hippocampus; also expressed in adipose tissue. Related references: PMID:34016966
- Glycoprotein with a predicted signal peptide and transmembrane helix, supporting its dual membrane-bound and secreted nature.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 45–167 of human FNDC4, covering a portion of the extracellular region; this may detect both secreted and membrane-bound forms.
- For IHC-P, consider optimized antigen retrieval and validate in tissues with known high expression, such as liver or brain.
- For ELISA, paired antibodies can be tested with recombinant FNDC4 protein; confirm cross-reactivity in mouse samples if required.
