Biological Background: FMRP Function and Localization
- FMRP (Fragile X messenger ribonucleoprotein 1), encoded by the FMR1 gene, is a multifunctional polyribosome-associated RNA-binding protein crucial for neuronal development and synaptic plasticity. It regulates alternative mRNA splicing, mRNA stability, dendritic transport, and local protein synthesis at synapses. - Acts as both a translational repressor and activator of target mRNAs. In synapses, it typically represses translation via the CYFIP1-EIF4E-FMR1 complex and by forming phase-separated ribonucleoprotein granules that inhibit translation. However, upon metabotropic glutamate receptor (mGluR) stimulation, it can activate translation of a subset of dendritic mRNAs. - Binds to specific RNA motifs including G-quadruplex structures, U-rich sequences, and the ACU[GU] and [AU]GGA consensus sequences, primarily within coding sequences and 3'UTRs of target mRNAs. - Localizes extensively in the cytoplasm, particularly at neuronal projections including dendrites, axons, dendritic spines, and growth cones. It is found in ribonucleoprotein granules, stress granules, and at synaptic sites (both pre- and postsynaptic). Nuclear localization includes the nucleolus, Cajal bodies, and centromere. It also associates with the cell membrane.
- Post-translationally modified by acetylation, methylation, and phosphorylation (including phase-separation-promoting phosphorylation). Ubiquitination is also noted. Mutations in FMR1 cause fragile X syndrome (intellectual disability) and premature ovarian failure; the protein is implicated in neurodegeneration.
- Exhibits tissue-specific expression, mainly in brain, cerebellum, testis, and epithelial tissues. At the protein level, detected in neurons, Purkinje cells, spermatogonias, and lymphocytes.

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide covering residues 1–100 of human FMRP (Q06787); thus, the antibody is expected to recognize the N-terminal region. For western blot (WB), a band at ~71 kDa is predicted; consider using lysates from brain or testis (high endogenous expression) as positive controls.
- For immunohistochemistry (IHC-P), optimal staining may require antigen retrieval and careful titration; validation in human, mouse, or rat neuronal tissues is recommended.
- In ELISA, the peptide immunogen can serve as a standard for quantitative assays; paired with an appropriate detection antibody, this clone may be suitable for sandwich ELISA development.
