Biological Background: FLT3 Function and Localization
- FLT3 (Fms-like tyrosine kinase 3), also known as CD135, FLK2, STK1, is a receptor-type tyrosine-protein kinase that acts as a cell-surface receptor for the cytokine FLT3LG, playing a central role in the regulation of differentiation, proliferation, and survival of hematopoietic progenitor cells and dendritic cells.
- Upon ligand binding, FLT3 activates multiple downstream signaling pathways, including phosphorylation of SHC1 and AKT1, activation of mTOR, RAS signaling, and MAPK/ERK cascades, thereby promoting cell growth and inhibiting apoptosis.
- FLT3 is a member of the class III receptor tyrosine kinase family, characterized by an extracellular region with five immunoglobulin-like domains, a single transmembrane helix, and an intracellular kinase domain; alternative splicing generates isoforms with distinct properties.
