Biological Background: MADS-box protein FLOWERING LOCUS C (FLC) Function and Localization
- FLC is a MADS-box transcription factor that functions as a central repressor of the transition from vegetative to reproductive growth in Arabidopsis thaliana.
- It integrates signals from the FRIGIDA-dependent autonomous pathway and the vernalization pathway to delay flowering, primarily by repressing the expression of the floral integrator SUPPRESSOR OF OVEREXPRESSION OF CONSTANS 1 (SOC1).
- At elevated temperatures (e.g., 29°C), FLC is maintained at high levels in a JMJ30/JMJ32-dependent manner to prevent extreme precocious flowering. Related references: PMID:25267112
- FLC localizes to the nucleus, consistent with its role as a transcription factor.
- The protein is highly expressed in the vegetative apex and root tissue, with lower expression in leaves and stems, and is absent from young inflorescence tissues. Before fertilization, it is expressed in ovules but not in pollen or stamens; after vernalization, it is undetectable in ovules.
- Alternative splicing of FLC produces multiple isoforms, which may contribute to regulatory diversity in flowering time control.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 60-196 of FLC, a region conserved among known isoforms; thus, the antibody may detect multiple FLC variants. Validate specificity using recombinant protein controls.
- For Western blot, expect a band at approximately 22 kDa under reducing conditions. Include positive (e.g., tissue with high FLC expression) and negative (e.g., vernalized tissue) controls to confirm detection.
- For ELISA, optimize coating and detection conditions using the recombinant immunogen or full-length FLC as standard.
- Because FLC expression is influenced by developmental stage, vernalization, and temperature, carefully design sample collection to capture the desired expression state.
