Biological Background: Folliculin Function and Localization
- FLCN is a multi-functional protein encoded by the FLCN gene, also known as BHD (Birt-Hogg-Dubé syndrome) protein, and acts as a tumor suppressor.
- It functions as a GTPase-activating protein (GAP) for RagC/RRAGC and RagD/RRAGD, stimulating GTP hydrolysis to activate mTORC1 in response to amino acid availability. Related references: PMID:24095279 PMID:31672913 PMID:31704029
- FLCN plays a key role in the non-canonical mTORC1 signaling pathway controlling the subcellular localization of MiT/TFE transcription factors TFEB and TFE3, thereby regulating lysosomal biogenesis and autophagy. Related references: PMID:17028174 PMID:18663353 PMID:21209915 PMID:24081491 PMID:31672913
- In low amino acid conditions, the lysosomal folliculin complex (LFC) inhibits FLCN's GAP activity, leading to mTORC1 inactivation and nuclear translocation of TFEB/TFE3; upon amino acid restimulation, the complex disassembles, restoring mTORC1 activation. Related references: PMID:31672913 PMID:32612235
- FLCN indirectly acts as a positive regulator of Wnt signaling by promoting mTOR-dependent cytoplasmic retention of TFE3, and it is required for the exit of hematopoietic stem cells from pluripotency. Related references: PMID:31272105 PMID:30733432
- It regulates glycolysis by binding to lactate dehydrogenase LDHA and acting as an uncompetitive inhibitor. Related references: PMID:34381247
- In response to flow stress, FLCN regulates STK11/LKB1 accumulation and mTORC1 activation through primary cilia by recruiting STK11/LKB1 to cilia for AMPK activation. Related references: PMID:27072130
- Following phosphorylation by ULK1, FLCN interacts with GABARAP and promotes autophagy; it is also required for starvation-induced perinuclear clustering of lysosomes via RILP-RAB34. Related references: PMID:25126726 PMID:27113757
- Subcellular localization includes lysosome membrane, cytosol, primary cilium, centrosome, spindle, and nucleus. It is expressed in most tissues, including skin, lung, kidney, heart, testis, and stomach. FLCN is associated with Birt-Hogg-Dubé syndrome and acts as a tumor suppressor.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 341–579, located in the C-terminal region; therefore, the antibody may recognize full-length FLCN and potentially some splice variants.
- For Western blot, the predicted molecular weight is ~64 kDa; include appropriate positive and negative controls to confirm specificity.
- In IF/ICC, consider permeabilization conditions optimized for lysosomal and cytoplasmic targets; co-staining with organelle markers can help confirm localization.
- For ELISA, the polyclonal nature provides broad epitope coverage; titrate the antibody to determine optimal working concentration.
- Cross-reactivity is confirmed for human, mouse, and rat, but validation in your specific model system is recommended.
