  • FKBP52/FKBP4, also known as FKBP51, Hsp56, p59, or HBI, is a member of the immunophilin family with peptidyl-prolyl cis-trans isomerase (PPIase) and co-chaperone activities.
  • It serves as a component of steroid receptor heterocomplexes by interacting with heat-shock protein 90 (HSP90), facilitating intracellular trafficking of steroid hormone receptors between the cytoplasm and nucleus.
  • The protein is widely expressed and localizes to multiple cellular compartments, including the cytoplasm, cytosol, mitochondrion, nucleus, cytoskeleton, and neuronal axons.
  • FKBP52/FKBP4 regulates neuronal growth cone dynamics through its isomerase activity by modulating TRPC1 channel opening, and also controls microtubule assembly by inhibiting MAPT/TAU.
  • It may have a protective role against oxidative stress in mitochondria.
