Biological Background: FKBP38/FKBP8 Function and Localization

  • FKBP8, also known as FKBP38, hFKBP38, FKBP-38, FK506-binding protein 8, FKBPR38, or Rotamase, is a peptidyl-prolyl cis-trans isomerase (PPiase) belonging to the immunophilin family. It contains TPR repeats and a putative transmembrane helix.
  • The enzyme is constitutively inactive as a PPiase and becomes active upon binding to calmodulin and calcium. It functions as a chaperone for BCL2, targeting BCL2 to mitochondria and modulating its phosphorylation state, thereby likely regulating apoptosis.

  • The protein localizes to mitochondrial membranes and is an integral membrane protein.
