  • FKBP12, also known as FKBP1A, FKBP1, or calstabin-1, is a 12 kDa FK506-binding protein that functions as a peptidyl-prolyl cis-trans isomerase (PPIase), accelerating protein folding by catalyzing the cis-trans isomerization of proline imidic peptide bonds in oligopeptides.
  • It plays a key role in TGF-beta signaling by maintaining the type I receptor TGFBR1 in an inactive conformation in the absence of ligand, thereby preventing premature receptor activation.
  • FKBP12 recruits SMAD7 to the activin receptor ACVR1B, inhibiting the association of SMAD2 and SMAD3 with the receptor complex and thus blocking activin signal transduction.
  • The protein may modulate the activity of the ryanodine receptor (RYR1) calcium release channel, implicating it in calcium homeostasis.
