Biological Background: FKBP10 Function and Localization
- FKBP10 encodes a 65 kDa FK506-binding protein (FKBP65), a peptidyl-prolyl cis-trans isomerase (PPIase) that accelerates protein folding by catalyzing the isomerization of proline peptide bonds.
- It belongs to the FKBP immunophilin family and contains multiple FKBP-type peptidyl-prolyl cis-trans isomerase domains.
- FKBP10 is localized to the endoplasmic reticulum lumen, where it functions as a molecular chaperone involved in collagen biosynthesis and protein quality control.
- The protein is a glycoprotein and calcium-binding metalloprotein, with post-translational modifications including phosphorylation (Keywords: Glycoprotein, Calcium, Metal-binding, Phosphoprotein).
- Alternative splicing generates multiple transcript variants, potentially contributing to functional diversity.
- Mutations in FKBP10 are associated with osteogenesis imperfecta, a genetic disorder characterized by bone fragility and connective tissue abnormalities.
Experimental Guidance and Technical Tips
- The antibody was raised against recombinant protein; it may recognize both denatured and native FKBP10, suitable for western blotting, immunofluorescence, and ELISA.
- For western blotting, a band at approximately 64 kDa is expected; consider loading appropriate molecular weight markers and using a suitable blocking buffer.
- When performing immunofluorescence, permeabilization is required to detect this endoplasmic reticulum-luminal protein; co-stain with an ER marker to confirm localization.
- Validation in cell lines or tissues expressing FKBP10, such as osteoblasts or fibroblasts, is recommended to confirm specificity.
