Biological Background: FILIP1L Function and Localization
- FILIP1L (Filamin A-interacting protein 1-like), also known as DOC1, GIP90, and GIP130, is encoded by the FILIP1L gene. Multiple isoforms exist due to alternative splicing, including 90 kDa and 130 kDa variants.
- Acts as a regulator of antiangiogenic activity in endothelial cells; overexpression inhibits cell proliferation and migration and increases apoptosis.
- When expressed in tumor-associated vasculature, FILIP1L inhibits melanoma growth.
- Subcellular localization includes the cytoplasm, membrane, and nucleus.
- Expressed in endothelial cells, colon, and ovarian epithelial cells. Notably down-regulated in ovarian cancer and strongly expressed in colon cancer stroma and vasculature.
- Classified as a phosphoprotein, suggesting regulation by phosphorylation.
- Contains coiled coil domains and has been identified through proteomics.
Experimental Guidance and Technical Tips
- The immunogen (aa 1–200) covers the N-terminal region; consider that alternative isoforms lacking this region may not be detected.
- For western blotting, a band around 130 kDa is expected based on the full-length protein; smaller bands may correspond to isoforms or cleavage products.
- Validate antibody specificity in the relevant sample system using positive and negative controls (e.g., tissues with known FILIP1L expression).
- For ELISA applications, the recombinant immunogen can serve as a positive control for assay development.
