Biological Background: Filamin-C Function and Localization

  • Filamin-C (FLNC), also known as ABP-280-like protein, gamma-filamin, or filamin-2, is a muscle-specific member of the filamin family that cross-links actin filaments and provides structural integrity at the Z-lines of sarcomeres.
  • This protein plays a central role in sarcomere assembly and organization, which is critical for normal myogenesis and muscle function.

  • FLNC functions as a large actin-cross-linking protein with structural roles at the Z-lines of muscle cells, and it may also participate in reorganizing the actin cytoskeleton in response to signaling events (By similarity).
  • Encoded by the FLNC gene, which belongs to the filamin gene family (including FLNA and FLNB), FLNC is alternatively spliced and subject to post-translational modifications such as phosphorylation, methylation, and ubiquitination.
  • Subcellularly, FLNC localizes to the cytoplasm, specifically associated with the cytoskeleton, and is prominently found in the myofibril sarcomere Z-line; it can also associate with cell membranes.
  • Tissue expression is highly enriched in striated muscles, with weaker expression in thyroid, fetal brain, fetal lung, retina, spinal cord, and bone marrow; it is not detected in testis, pancreas, adrenal gland, placenta, liver, or kidney.
  • Mutations in FLNC are associated with various cardiomyopathies and myofibrillar myopathies, highlighting its essential role in cardiac and skeletal muscle stability.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 2160–2340 within the C-terminal region of human FLNC. This sequence may be conserved across isoforms and species, as supported by the antibody's cross-reactivity with mouse.
  • For Western blot, FLNC is a large protein (~291 kDa); using low-percentage (e.g., 6–8%) SDS-PAGE gels, extended transfer times, and a suitable molecular weight marker is recommended. Confirm band specificity against the predicted size in relevant tissue or cell lysates.
  • In immunofluorescence/ICC applications, the antibody should label myofibrillar structures, particularly Z-lines, in striated muscle cells. Co-staining with sarcomeric markers (e.g., α-actinin) can help validate localization.
  • For ELISA, standard direct or indirect formats may be applied; we suggest titrating the antibody and testing with recombinant FLNC protein or lysates from muscle tissues.
