Biological Background: Filamin-B Function and Localization
- Filamin-B (FLNB) is an actin-binding protein that crosslinks actin filaments and connects the cell membrane to the cytoskeleton.
- It promotes orthogonal branching of actin filaments and links actin filaments to membrane glycoproteins, anchoring various transmembrane proteins to the actin cytoskeleton.
- Interaction with filamin A (FLNA) may facilitate neuroblast migration from the ventricular zone to the cortical plate.
- Various isoforms (1–6) exist via alternative splicing; isoform 6 has been shown to accelerate muscle differentiation in vitro.
- Subcellular localization includes cytoplasm, cell cortex, stress fibers, and Z-lines of sarcomeres in muscle cells.
- Ubiquitously expressed with isoform-specific distribution: isoform 1 is predominant in many tissues, while others are enriched in heart, skeletal muscle, and leukocytes.
- Keywords: Acetylation, Actin-binding, Alternative splicing, Differentiation, Isopeptide bond, Myogenesis, Phosphoprotein, Ubl conjugation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1686–1785 of human FLNB; consider this region when designing validation experiments or assessing isoform recognition.
- For WB, the predicted molecular weight is 278 kDa; use a low-percentage SDS-PAGE gel and extended transfer times to ensure efficient blotting of this high-molecular-weight protein.
- The antibody is applicable for IHC-P, IF/ICC, and ELISA; we recommend titrating the antibody and including appropriate positive and negative controls for each assay.
- Cross-reactivity with mouse and rat is reported based on sequence homology; verify antibody specificity in your target species samples if precise quantification is required.
