Biological Background: Filaggrin Function and Localization
- Filaggrin (FLG) is a large, highly phosphorylated histidine-rich protein that serves as a key structural component in epidermal differentiation.
- It aggregates keratin intermediate filaments into dense macrofibrils and promotes disulfide-bond formation, contributing to the mechanical strength and barrier function of the skin.
- FLG is initially synthesized as profilaggrin and stored in keratohyalin granules; upon terminal differentiation, it is proteolytically processed into functional filaggrin peptides.
- Subcellularly, filaggrin localizes to cytoplasmic keratohyalin granules within granular layer keratinocytes.
- Tissue expression is predominantly in the skin (stratum granulosum), but also detected in thymus, stomach, tonsils, testis, placenta, kidney, pancreas, mammary gland, bladder, thyroid, salivary gland, and trachea, with absence in heart, brain, liver, lung, bone marrow, small intestine, spleen, prostate, colon, and adrenal gland. - The immunogen corresponds to the N-terminal region (aa 1-92) of human FLG, which may recognize the profilaggrin precursor as well as processed filaggrin. Consider including protease inhibitors during sample preparation to prevent degradation.
- For Western blot, human skin keratinocyte lysates (e.g., HaCaT cells under calcium-induced differentiation) or tissue extracts from skin can be used to detect the full-length ~435 kDa profilaggrin and its proteolytic fragments. Validate the antibody in the relevant sample system.
- This antibody is suitable for ELISA; pairing with a matched detection antibody may enable sensitive sandwich immunoassays for filaggrin quantification in biological fluids or cell supernatants.
- Cross-reactivity with mouse and rat has been confirmed; ensure optimization of species-specific protocols when using non-human samples.
