Biological Background: Fibroblast Activation Protein Alpha (FAP) Function and Localization
- FAP, also known as seprase and SIMP, is a cell-surface glycoprotein serine protease.
- Its endopeptidase and dipeptidyl peptidase activities target substrates like alpha-2-antiplasmin, SPRY2, and neuropeptides (NPY, PYY, substance P), with preference for Ala/Ser-Gly-Pro-Ser/Asn/Ala consensus sequences.

Related references: PMID:10347120, PMID:10455171, PMID:12376466 - In tumors, FAP promotes glioma invasion by degrading brevican and enhances angiogenesis, collagen degradation, and immune evasion, supporting malignant progression.
- Subcellularly, FAP is found on the cell membrane, lamellipodia, invadopodia, and ruffles; it can also be secreted or present in the cytoplasm.
- Tissue expression is restricted to activated fibroblasts (e.g., wound healing, cancer stroma, cirrhotic liver), glioma cells, and adipose tissue, but absent in normal liver.
- The FAP gene produces at least two isoforms via alternative splicing, both expressed in melanoma, carcinoma, and fibroblast cell lines.
Experimental Guidance and Technical Tips
- For Western blot, the predicted molecular weight of FAP is ~88 kDa; use appropriate gel percentages (e.g., 8–10% SDS-PAGE) and confirm band specificity with relevant controls.
- In immunofluorescence/ICC, permeabilization steps are essential to detect membrane-localized FAP; validate staining in positive cell lines such as activated fibroblasts or cancer cells.
- For ELISA applications, this polyclonal antibody may serve as a capture or detection reagent; pairing optimization and appropriate blocking are recommended.
- Given the polyclonal nature and near-full-length immunogen, the antibody likely recognizes multiple epitopes, which can be advantageous for detecting variants across human, mouse, and rat.
- Verify cross-reactivity with mouse and rat samples using species-specific positive controls, as expression levels may vary.
- Consider that soluble FAP is present in some biological fluids, which could compete with membrane-bound forms in certain assays.
