Biological Background: Fibrillarin/U3 RNP Function and Localization
- Fibrillarin is an rRNA 2'-O-methyltransferase, also known as 34 kDa nucleolar scleroderma antigen, histone-glutamine methyltransferase, and U6 snRNA 2'-O-methyltransferase fibrillarin.
- It catalyzes site-specific 2'-hydroxyl methylation of ribose moieties in pre-ribosomal RNA, guided by box C/D snoRNAs. Related references: PMID:30540930
- Functions as a protein methyltransferase by methylating histone H2A at glutamine 105 (H2AQ104me), which impairs FACT complex binding at the 35S ribosomal DNA locus. Related references: PMID:24352239, PMID:30540930
- Methylates U6 snRNA at specific 2'-O sites within box C/D RNP complexes, a modification essential for mRNA splicing fidelity. Related references: PMID:32017898
- Acts as an integral component of the small subunit (SSU) processome, the first precursor of the small ribosomal subunit, and is critical for ribosome biogenesis. Related references: PMID:34516797
- Predominantly localizes to the nucleolus and nucleoplasm of the nucleus.
- Post-translational modifications include acetylation, phosphorylation, isopeptide bond formation, and ubiquitin conjugation; it is also methylated itself and contains RNA-binding domains.
Experimental Guidance and Technical Tips
- For western blot, a band approximately 34 kDa is expected; use appropriate molecular weight markers for confirmation.
- The immunogen region (amino acids 222-321) shows conservation across human, mouse, and rat, supporting the observed cross-reactivity; validate performance in your specific sample system.
- In immunofluorescence or ICC, nucleolar localization should be evident; optimize fixation and permeabilization conditions for nuclear target detection.
- For immunoprecipitation, verify antibody compatibility with your extraction buffer and consider including a no-antibody control.
- For ELISA applications, assess coating conditions and use a known positive control such as the immunogen peptide to establish assay parameters.
