Biological Background: FHL2 Function and Localization

  • FHL2 (Four and a half LIM domains protein 2), also known as DRAL, AAG11, SLIM3, is a LIM domain protein that functions as a molecular transmitter linking various signaling pathways to transcriptional regulation.
  • It negatively regulates the transcriptional repressor E4F1, potentially influencing cell growth.
  • FHL2 inhibits FOXO1 transcriptional activity and its apoptotic function by enhancing FOXO1 interaction with SIRT1 and subsequent deacetylation.
  • Negatively regulates the calcineurin/NFAT signaling pathway in cardiomyocytes. Related references: PMID:28717008
  • The protein contains LIM domains, which are zinc-finger motifs mediating protein-protein interactions, and is subject to alternative splicing, phosphorylation, and ubiquitination.
  • It localizes to the cytoplasm, nucleus, and specifically to the Z line of sarcomeres in muscle cells, reflecting its role in muscle biology.
  • Tissue specificity: Predominantly expressed in skeletal muscle and heart.

Experimental Guidance and Technical Tips

  • The recombinant immunogen spans amino acids 50–260 of human FHL2, encompassing key LIM domains; this region may ensure specificity for FHL2 across reactive species (human, mouse, rat).
  • For Western blot, expect a band at approximately 32 kDa. Due to potential phosphorylation and other modifications, minor shifts or additional bands may be observed; validate using positive controls from skeletal muscle or heart tissues.
  • For ELISA, validate the antibody’s reactivity with the target protein in the relevant sample matrix and consider a blocking strategy to reduce non-specific binding.
  • If cross-reactivity with other FHL family members is a concern, verify specificity by including appropriate controls or using knockdown/knockout models.
