Biological Background: Fumarate Hydratase Function and Localization
- FH catalyzes the reversible stereospecific interconversion of fumarate to L-malate, a reaction central to mitochondrial energy production and cytosolic metabolic pathways. Related references: PMID:30761759
- In the mitochondrial tricarboxylic acid (TCA) cycle, FH hydrates fumarate to L-malate, contributing to NADH generation. In the cytosol, it dehydrates L-malate to fumarate, supporting the urea cycle and arginine catabolism.
- Alternative translation initiation produces mitochondrial and cytosolic isoforms, which may exhibit opposite catalytic preferences according to evidence from other species (Probable).
- FH participates in DNA double-strand break repair by translocating to the nucleus after DNA damage and promoting non-homologous end joining (NHEJ). It catalyzes fumarate formation, which inhibits the KDM2B histone demethylase, leading to elevated H3K36me2 and facilitating repair protein assembly. Related references: PMID:20231875 PMID:26237645
- Subcellular localization spans mitochondria, cytoplasm, nucleus, and chromosome; nuclear accumulation is induced by genotoxic stress and phosphorylation by PRKDC.
- FH acts as a tumor suppressor; germline mutations are linked to hereditary leiomyomatosis and renal cell cancer (HLRCC) and fumarate hydratase deficiency.
- Post-translational modifications, including acetylation and phosphorylation, regulate FH activity and intracellular trafficking.
- FH is expressed in red blood cells, and reduced cytoplasmic levels have been observed in patients with hereditary non-spherocytic hemolytic anemia.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 45–510 of human FH, encompassing the majority of the protein sequence. This design likely detects both mitochondrial and cytosolic isoforms.
- For Western blot analysis, the predicted molecular weight is approximately 55 kDa. Multiple bands may appear due to isoform differences or post-translational modifications; validate specificity using appropriate controls.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and fixation protocols. Positive and negative tissue controls expressing FH at known levels are recommended.
- In ELISA applications, consider using recombinant FH protein as a standard for quantitative assays and verify cross-reactivity with homologous proteins from other species.
