Biological Background: FH (Fumarate Hydratase) Function and Localization

  • FH catalyzes the reversible hydration of fumarate to L-malate in the TCA cycle, a key step in mitochondrial energy production (PubMed:30761759). Related references: PMID:30761759

  • Cytosolic FH isoforms participate in the urea cycle and arginine metabolism, producing fumarate as a by-product (By similarity).

  • FH is annotated as a tumor suppressor and disease variant protein; mutations are linked to hereditary cancer syndromes such as HLRCC.

  • In response to DNA damage, FH is phosphorylated by PRKDC and translocates to the nucleus, where it promotes NHEJ repair by inhibiting KDM2B demethylase, leading to increased H3K36me2 at DSBs (PubMed:20231875, PubMed:26237645). Related references: PMID:20231875, PMID:26237645

  • FH localizes primarily to mitochondria but also to the cytosol; nuclear accumulation occurs after DNA damage (PubMed:26237645).

  • Alternative initiation produces different isoforms that may favor either hydration or dehydration reactions (Probable).

  • FH is expressed in red blood cells and is underexpressed in patients with hereditary non-spherocytic hemolytic anemia of unknown etiology.

  • Post-translational modifications include acetylation and phosphorylation; PRKDC-mediated phosphorylation at DSBs is critical for its DNA repair role.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 50–150 of human FH; consider using positive controls such as HeLa or HEK293T lysates for WB.
  • For Western blotting, a specific band at ~55 kDa should be observed; use appropriate loading controls and molecular weight markers.
  • Subcellular fractionation may improve detection of mitochondrial or nuclear FH pools.
  • For ELISA, optimize coating antigen and antibody titration to achieve best signal-to-noise ratios.
  • Cross-reactivity with mouse and rat FH allows multispecies studies; validate in your experimental system.
  • Pre-adsorption with the immunizing peptide can confirm band specificity.
