Biological Background: FGFR4 Function and Localization

  • FGFR4 is a fibroblast growth factor receptor and tyrosine-protein kinase that acts as a cell-surface receptor for fibroblast growth factors, regulating cell proliferation, differentiation, and migration.
  • It plays a critical role in metabolic regulation, including lipid metabolism, bile acid biosynthesis, glucose uptake, vitamin D metabolism, and phosphate homeostasis. FGFR4 is required for the normal down-regulation of CYP7A1, the rate-limiting enzyme in bile acid synthesis, in response to FGF19.
  • Ligand binding triggers activation of multiple signaling cascades: phosphorylation of PLCG1 generates diacylglycerol and inositol 1,4,5-trisphosphate; phosphorylation of FRS2 recruits GRB2, GAB1, PIK3R1, and SOS1, leading to activation of RAS, MAPK1/ERK2, MAPK3/ERK1, and the AKT1 pathway.
  • FGFR4 signaling is down-regulated by receptor internalization and degradation, a process promoted by SRC-dependent phosphorylation of the matrix protease MMP14 and its subsequent lysosomal degradation.
  • Subcellular localization includes the cell membrane, endosomes, endoplasmic reticulum, and a secreted form.
  • It is expressed in gastrointestinal epithelial cells, pancreas, and gastric and pancreatic cancer cell lines.

  • The immunogen corresponds to amino acids 22–150 in the extracellular domain; the resulting polyclonal antibody may preferentially recognize epitopes in the N-terminal region, which is useful for detecting cell surface FGFR4.
  • In Western blotting (WB), a band around 88 kDa is expected; however, glycosylation and other modifications may cause slight shifts in apparent molecular weight. Validate the antibody in your specific sample system.
  • For ELISA applications, consider pairing this antibody with a suitable capture antibody if developing a sandwich format. Perform cross-reactivity testing against mouse and rat samples as indicated, and include appropriate negative controls.
