Biological Background: FGFR3 Function and Localization

  • FGFR3 (Fibroblast Growth Factor Receptor 3) is a receptor tyrosine kinase that binds fibroblast growth factors, thereby regulating key cellular processes such as proliferation, differentiation, and apoptosis.
  • It plays an essential role in chondrocyte differentiation and skeletal development; FGFR3 is required for normal bone mineralization by osteoblasts and regulates both osteogenesis and postnatal bone remodeling.
  • Upon ligand binding, FGFR3 triggers multiple intracellular signaling cascades, including the MAPK/ERK (via phosphorylation of FRS2 and recruitment of GRB2, SOS1) and AKT1 pathways, as well as PLCG1-mediated production of second messengers diacylglycerol and inositol 1,4,5-trisphosphate.
  • FGFR3 is also involved in the regulation of vitamin D metabolism.
  • Subcellular localization includes the cell membrane, cytoplasmic vesicles, and the endoplasmic reticulum; a secreted isoform (isoform 3) exists that retains FGF-binding capacity and may modulate signaling.
  • Tissue expression is highest in brain, kidney, and testis; during fetal development, strong expression is observed in kidney, lung, small intestine, and brain. Fibroblastic cells express both isoform 1 and isoform 2, while epithelial cells solely express isoform 2.
  • Mutations leading to constitutive activation of FGFR3 are associated with craniosynostosis, various forms of dwarfism, deafness, and cancer. Overexpression or aberrant activation promotes phosphorylation of STAT1, STAT5A/B, and SHP2, driving oncogenic signaling.

Experimental Guidance and Technical Tips

  • The immunogen targets amino acids 700–806 within the intracellular tyrosine kinase domain of FGFR3; thus, the antibody detects the cytoplasmic portion of the receptor. For IHC-P and IF/ICC, ensure adequate permeabilization to allow access to this epitope.
  • In WB, the antibody is expected to recognize FGFR3 at approximately 88 kDa. Include appropriate positive (e.g., FGFR3-expressing cell lines) and negative controls to validate specificity in your experimental system.
  • For ELISA applications, the antibody can be used as capture or detection reagent; optimization of coating concentrations and pairing with other FGFR3 antibodies is recommended.
