Biological Background: FGF1 Function and Localization
- Fibroblast growth factor 1 (FGF1), also known as acidic FGF (aFGF), endothelial cell growth factor (ECGF), and heparin-binding growth factor 1 (HBGF-1), is a member of the fibroblast growth factor family.
- FGF1 acts as a potent mitogen and plays an important role in the regulation of cell survival, cell division, angiogenesis, cell differentiation, and cell migration.
- It functions as a ligand for FGFR1 and integrins (ITGAV:ITGB3), triggering downstream signaling. FGF1 induces the phosphorylation and activation of FGFR1, FRS2, MAPK3/ERK1, MAPK1/ERK2, and AKT1. Related references: PMID:18441324, PMID:20422052

- Subcellularly, FGF1 is found in the cytoplasm, cell cortex, cytosol, and nucleus, and it is also secreted.

- The immunogen corresponds to amino acids 56-155 of human FGF1, covering the mature protein sequence; consider using recombinant full-length or mature FGF1 as a positive control.
- For Western blotting (WB), the predicted molecular weight is ~17 kDa; optimize loading and transfer conditions for low-molecular-weight proteins.
- FGF1 is heparin-binding; in immunoassays, the inclusion of heparin or low-ionic-strength buffers may influence antibody-antigen interactions and should be assessed.
- When performing immunohistochemistry (IHC-P) or immunofluorescence (IF/ICC), validate staining in high-expression tissues such as kidney or brain.
- For ELISA, titrate the antibody against recombinant FGF1 standards and confirm linearity and sensitivity in the relevant sample matrix.
