Biological Background: FGF1 Function and Localization
- FGF1 is a fibroblast growth factor family member, also known as acidic FGF (aFGF), endothelial cell growth factor (ECGF), and heparin-binding growth factor 1 (HBGF-1).
- It regulates cell survival, division, angiogenesis, differentiation, and migration, acting as a potent mitogen in vitro.
- FGF1 binds to FGFR1 with heparin, leading to receptor dimerization, autophosphorylation, and activation of downstream signaling cascades.
- It induces phosphorylation and activation of FGFR1, FRS2, MAPK3/ERK1, MAPK1/ERK2, and AKT1. Related references: PMID:18441324 PMID:20422052
- FGF1 can induce angiogenesis, a process crucial for tissue repair and tumor growth. Related references: PMID:23469107
- FGF1 also binds to integrin ITGAV:ITGB3, forming a ternary complex with integrin and FGFR1, essential for signaling.
- Subcellularly, FGF1 localizes to the secreted space, cytoplasm, cell cortex, cytosol, and nucleus.
- It is predominantly expressed in kidney and brain, with lower levels in heart and skeletal muscle; post-translational modifications include acetylation and phosphorylation, and alternative splicing yields multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 16-155 of human FGF1, covering the mature protein, which may confer broad reactivity with the native form.
- For WB, a band at approximately 17 kDa is expected; however, post-translational modifications may shift the observed molecular weight.
- In IHC-P and IF/ICC, consider using appropriate fixation and antigen retrieval methods; validate in tissues known to express FGF1 such as kidney or brain.
- ELISA performance may vary; consider sandwich ELISA development with a capture/detection pair.
- Cross-reactivity with mouse and rat FGF1 is indicated; verify specificity in the relevant species context.
